新疆阿魏蘑菇提取物体外抗肿瘤实验研究

摘要: 目的: 探讨新疆阿魏蘑菇提取物对体外培养的肿瘤细胞生长的抑制作用。方法:不同剂量阿魏蘑菇的水提物和醇提物(10 - 1 g/ ml、10 - 2 g/ ml、10 - 3 g/ ml、10 - 4 g/ ml、10 - 5 g/ ml、10 - 6 g/ ml) 与肿瘤细胞共培养24 h 后,采用结晶紫染色及Bradford 法测定人肝癌细胞株(Q3) 、人胃癌细胞株(MGC2803) 、人宫颈癌细胞株(Hela) 、小鼠肺腺瘤细胞株(SPC2A21) 的细胞存活量及蛋白质合成。以大鼠正常肝细胞(BRL) 为正常对照细胞株。结果:阿魏蘑菇水提物、醇提物10 - 1 g/ ml 剂量组对各类型肿瘤细胞的生长及蛋白合成均有明显抑制作用;醇提物不同剂量组对肿瘤细胞生长及蛋白合成可产生不同程度的影响,对正常肝细胞的存活量无影响,但可促进肝细胞的蛋白合成。结论:阿魏蘑菇提取物的抗肿瘤作用可能与其抑制肿瘤细胞生长及蛋白合成有关。

Abstract: Purpose : To study the effect of the extract of Pleurotas sapidus on 4 tumor cell lines ( human hepatoma cell Q3 , human gastric cancer cell MGC-803 , human cervical carcinoma Hela cell , murine lung adenocar-cinoma cell SPC-A-1) . Methods : The survival cells and protein synthesis of tumor cell lines treated for 24 h with 10 - 1 g/ ml~10 - 6 g/ ml of the water and ethanol extracts from Pleurotas sapidus were determined by crystal violet staining and by Bradford assays. Results : ①There was a significant growth inhibition of tbe cell survival and protein synthesis of the cell lines at a dose of 10 - 1 g/ ml of the water and ethanol extracts ; ②The growth inhibition was found in different dose groups of ethanol extract . In the normal liver cells , the cell survival was not affected and the protein synthesis increased. Conclusion : The inhibition of the tumor cell lines with the extracts fom Pleurotas Sapidus may be related to the inhibition of cell growth and protein synthesis.
